1) Is the domain is https , is good to set Enable on Protocol both or https ?
2) CDN Domain , what is it better , multiple subdomains or one ?

1) The 'Enable on Protocol' both or https only makes a difference if you serve parts of your website through https and parts through http.

2) Depends on many factors. In most cases using one is fine. But you might want to serve images from one domain and css/js files from another. Maybe even a different cdn service.
Or you have video files that you want to serve from a separate video-specialized cdn service.

First things first:
Make sure your cdn urls are actually working.

So say you have a file on your website:

mydomain.com/templates/mytemplate/css/template.min.css
Then this url should be working:
subdomain.cdndomain.com/templates/mytemplate/css/template.min.css
If it isn't, then there is something wrong with the setup at your CDN provider.

See the previous reply.
First make sure the CDN urls are working.

if you want to set a subdomain goes
subdomain.cdndomain.com/
not
subdomain.domain.com/
?

That is something you should set up at your CDN provider.
So again: do your CDN urls work? If not, then make sure that works first.
